一、背景描述：（番茄凝集素DyLight 649）
Lycopersicon esculentum (Tomato) Lectin (LEL, TL), DyLight 649, is a bright deep-red/far-red LEL-fluorophore conjugate.
The tomato lectin LEL is widely used as marker of blood vessels and microglial cells in rodents and in neuroscience research.
It is a valuable tool in studies of rodent tumor angiogenesis and the tracing of neovascular development in xenograft models and brain research.
LEL-DyLight 649 can be applied directly to tissue sections, or it can be introduced through intravascular perfusion for fluorescence detection.
番茄凝集素 （LEL， TL），DyLight 649，是一種明亮的深紅色/遠紅 LEL-熒光團偶聯物。
番茄凝集素 LEL 被廣泛用作嚙齒動物血管和小膠質細胞的標志物以及神經科學研究。
它是研究嚙齒動物腫瘤血管生成以及異種移植模型中新生血管發育追蹤和大腦研究的寶貴工具。
LEL-DyLight 649 可直接應用于組織切片，也可以通過血管內灌注引入熒光檢測。
LEL is a very stable single-subunit glycoprotein containing ～50 percent arabinose and galactose and may form multimeric aggregates in solution.
It is provided at a 1 mg/mL concentration in 10 mM HEPES, 0.15 M NaCl, pH 7.5, 0.08% sodium azide, 0.1 mM Ca2+. If a precipitate forms upon long-term storage, warm to 37°C.
LEL 是一種非常穩定的單亞基糖蛋白，含有約 50% 的阿拉伯糖和半乳糖，可在溶液中形成多聚體聚集體。
它以 1 mg/mL 濃度在 10 mM HEPES、0.15 M NaCl、pH 7.5、0.08% 、0.1 mM Ca2+ 中提供。如果長期儲存時形成沉淀物，請加熱至 37°C。

二、熒光檢測與染色問題（番茄凝集素DyLight 649）
（一）熒光參數
激發波長：655 nm，發射波長：670 nm（遠紅光），可用 Cy5/深紅濾光片 檢測。
染料結合量優化，幾乎不含游離熒光素，但仍需避光操作以防淬滅。
（二）常見問題與解決
低染色效率：
可能因反應溫度過低（需≥37°C）或時間不足，優化孵育條件。
高背景信號：
未充分沖洗血管（增加PBS洗滌次數）；
游離染料殘留（用層析柱純化或增加含 0.5% Tween-20的PBS 洗滌）；
非特異性結合（用 5% BSA 封閉組織切片）。
